Signaling Pathway for Butanol Production in Solventogenic Clostridium Bacteria

Diplomová práce se zabývá studiem signální dráhy produkce butanolu bakterií rodu Clostridium. V první části pojednává o modelování signálních drah pomocí metod systémové biologie. Navazuje popisem zisku dat pro tvorbu a úpravu modelů signálních drah s hlavním zaměřením na techniky pro zjištění genové exprese, produkce a fenotypu. Třetí sekcí je získání základního modelu signální dráhy zapojené do produkce butanolu u solventogenních klostridií. Posledním bodem a zároveň hlavním cílem je vytvoření dynamického modelu signální dráhy produkce butanolu kmene Clostridium beijerinckii NRRL B-598, jeho vyhodnocení pomocí statické a dynamické analýzy a srovnání s biologickými daty.

In this study, we bring the first dynamic model of signaling pathway for butanol production in sporulating, solvent-producing bacterium, Clostridium beijerinckii NRRL B-598. The model allows to study functions of individual genes involved in the production of solvents and acids. We used an open platform Cell Collective for designing the model and stored it under the name ‘Signaling Pathway for Butanol Production in Clostridium beijerinckii NRRL B-598’ version 1.0 (https://research.cellcollective.org/?dashboard=true#36604:1/signaling-pathway-for-butanol-production-inclostridium-beijerinckii-nrrl-b598/10) in publicly available repository of Cell Collective.

The impact of biofuel on the damage of ignition engines

The Bachelor´s Thesis evaluates the influence of currently used bio-fuels on the damage caused to spark-iqnition engines. The first part of the thesis defines bio-fuels, their characteristics and fabrication. The second part looks into problematics of spark-ignition engines, describes the process of injection and combustion of fuel. Moreover in the second part of the thesis are stated analyzed studies of the influence of bio-fuels, mostly ethanol and buthanol, on the damage caused to various parts of spark-iqnition engine, as well as the impact on the greenhouse gases production.
Possibilities of fermentation production of butanol as raw material for motor fuels - introductory study

The aim of this work was to map out the theoretical basics of acetone-butanol fermentation. In addition, it is to find available data for possibility of the industrial use of this process and to make conclusion for next procedure. The microorganisms producing butanol, conditions of fermentation and usable raw material were described. Batch, Fed-batch and Continuous fermentation processes are described in more detail from the point of view of productivity of process and yield. Available data was evaluated, but there was a lack of technological data to construct an operational system. Therefore, an experimental pilot plant system was constructed that allowed different types of microorganisms and substrates to be tested. The pilot plant system is for single stage Continuous fermentation with gas stripping product recovery with possibility of expansion to two-stage Continuous fermentation. As well the balance of this process was done.
Sorption Characteristics of PDMS in Ternary Systems Benzene (1) + Alcohols (2) + Ionic Liquid (3) and Corresponding Binary Subsystems

In this work the study of sorption characteristics of PDMS in ternary system benzene (1) + methanol or butanol (2) + ionic liquid (3) and corresponding binary subsystems at the temperature of 20°C is presented. The experimental data on total sorption were determined gravimetrically [3], the experimental data on preferential sorption were evaluated from the concentration change induced by an immersion of the membrane into the liquid mixture [3]. The volume changes of swollen membrane immersed in the studied liquid systems were determined from the dimensional changes of the membrane samples changes in X and Y axis dimensions were monitored directly in liquid by an optical method [1, 3] and membrane sheet thickness was measured by an Inductive Dial Indicator (Mahr, Germany).
